If by at least one black letter you mean literally only one blackletter and the rest BS this will absolutely matter and look very bad. Try to take two black letters a semester and if you do that feel free to slide in some BS classes. I am currently a rising 2L on the absolute ass-end of the top 10% (magna/Order of the Coif), like literally by 0.01. This is allowing me to put "top 10%" on my resume for pre-OCI and OCI, and I really want to graduate magna. So basically what I want to do (especially with grade inflation) is do mostly seminars/experiential stuff/externships and not do Admin, etc. (Still planning on doing at least one major black-letter class per semester plus Con Law 2/Fed Courts.)
If I apply to clerkships, it would be after a year or two of experience with a firm, so like 4-5 years from now. How much will it hurt me at that point to have not done more black letter classes?

100% depends by the judge and the circumstances. A friend of mine got an offer from a judge on the spot and another friend got an offer from the same judge weeks later. I had an offer within minutes from one judge, a few hours from another judge, and then have had nothing but radio silence for over three weeks at this point from another judge.
