In the DC Circuit, most of the judges hire students (or applicants who went 3L-->district court clerkship). You'll sometimes find DC COA clerks who worked for a year with a fellowship or at a firm first. It's rare to hear of a clerk with multiple years of firm experience.
I'm less knowledgeable about the Second Circuit but I believe it's roughly the same deal there.

I'd imagine that your odds for a COA gig would be slightly lower as a 4th year than as a 2L.
