Let's say you are mass mailing and emailing the HR of the firm. Should you also be applying through their online application as well?

No. Follow the firm's directions re: whether they want you to email your application or apply through their web portal. In addition, if you have a connection (partner or associate) at the firm, mention your application to that connection.

Well hold on—at biglaw spots, if you literally know a partner or associate (or at some firms, I think, even another staff member), you should absolutely get those people to initially pass your thing through. This might then generate a direct email asking you to submit. In my experience, If you do it any other way, you risk not getting your connection $, not to mention that you might end up in some black box portal reviewed for keywords only by non lawyers.
