Some details: career fair site asks for resume and cover letter (uploaded as one document), while regular process also asks for transcript, writing sample, etc. This is all public interest btw.
If you mean applying again later, then sure do it. If you mean contacting the employer outside the career fair and applying while simultaneously applying at the career fair, then don't do it.
I've had an experience where I didn't get an interview at a career fair, but then applied at another event and ended up with an interview and call back.
