Do you live within driving distance of your old school?
Maybe a polite stop by her office to say hello during her office hours could do the trick?
E-mail is really the worst way to ask for LORs. Even if you're on the other side of the globe, a phone call is much more effective.

I disagree about calling v. emailing (because it depends on your relationship and some professors don't want you calling out of the blue - they'd prefer to have a minute to remember who you are and review your resume/transcript) BUT in this case, I'd try calling because the reason you haven't received a response may be as simple as your email got caught in a spam filter or he/she is swamped with classes beginning. Don't push - if they don't feel comfortable writing, then you don't want that letter anyway, but I would offer to drive up and meet in person if it's a possibility.

If the prof doesn't answer, then yes, I'd seek out the back up that you mentioned. You'd be surprised, especially if you went to a large uni - a prof may be more willing to write a letter (even if they only knew you in a large class) because they understand how difficult it is to find recommenders.

If you attend a large university and were not able to cultivate a relationship with any of your professors, consider asking a T.A. or lecturer who may have worked more closely with you. Likewise, if you have been out of school for several years, ask an employer who knows you and your work well.
Is it possible for you to get a hold of your TA? They'd be more likely to remember you probably and may be better able or more willing to write you a letter of rec.
I probably wouldn't tell your prof that she's your backup. Just say something like "I really enjoyed your class most in undergrad and I feel I performed very well in it, so I would appreciate a letter from you if you wouldn't mind...." yadda yadda yadda.
