You need to find a way to bounce back strong. It is likely you will see this person at the reception desk when you walk in, so I'd try to loosen things up with a joke right off the bat.

I HAVE GOTTEN THIS QUESTION.
The actual question was (or at least was supposed to be, but got botched by the secretary) "would you like to speak to any LGBT/minority interviewers?" To which the appropriate response would be either (a) yes, I would like to speak to [one or the other], or (b) I don't mind (but I don't need to). The idea being if you're in one of those groups, you can ask about it.
As someone who says dumb shit on the reg, I'm lolling hard imagining the OP hanging up the phone, thinking for a second, and then closing his eyes as the realization of the fuckup comes flooding in.

I don't think it's so much about being worried that the gays are getting shunned at work. I think it's more about wondering if there are many gay people at the firm, whether they hang out, and maybe (if it's a secondary market) how the gay scene is in that market in general. This is a common ploy used by bigoted firms to weed out the LGBT applicants. Yours was the correct response. You've got the offer in the bag.

I got something similar but at the end of the question she said "or no preference." I was already a bit nervous and could almost see myself making this same mistake. Instead I just said no preference, echoing her question, that would be the correct answer if asked (assuming you don't have a preference).
I'd say the best solution here is to email the HR person immediately, saying that you realize your response could have been taken as offensive, apologizing, stating that it wasn't your intent and that you have no preference but would be happy to meet with either.
