If I obtain one of the two required LORs 1 to 2 years prior to submitting my applications, will that hurt my chance of admission?

Not at all! If anything, it's a good thing if you're a few years away from applying (because a professor would be able to write you a better letter now compared to two years after having a class).

Question stemming from this, but if I was to hypothetically defer (I've sent both my LOR's to 12 diff schools this cycle) would I be able to send it again to 12 next year?

Yeah, you can definitely re-use LORs and send them to the same schools again. However, when re-applying you want to make sure your application is different from the past year, and a new LOR is a (very small) way to make your application different, so it's up to you. At schools I re-applied to, I sent one LOR that they had already seen, and one new one. HTH
